No, not really. I've done several 24-hour comics and never had the drawing hand cramp up - more often than not my knees/legs/bum hurts. I think decades of drawing has given the hand some decent stamina.

I have a friend who is a professional musician as well as a comic artist. His hand doesn't wear out either, probably for the same reason.
